ABSTRACT

Due to the infinite proliferation, strong migration and loss of contact inhibition of tumor cells, tumor has become the most intractable diseases to be cured in the world. At present, the main treatments of tumor diseases are surgical resection, radiotherapy, chemotherapy, targeted-therapy and immunotherapy. Although these measures can inhibit or kill the tumor to a certain extent, they still cannot avoid adverse reactions and drug resistance. After thousands of years of clinical practice, traditional Chinese medicine (TCM) has the characteristics of good curative effect, few adverse reactions and significantly improving the quality of life in patients, which provides new ideas for the prevention and treatment of tumors. As an endemic and rare plant in China, Tetrastigma hemsleyanum has been listed in the 2015 edition of Zhejiang Provincial Processing Specification of TCM with the effects of heat-clearing and detoxification, detumescence and analgesia, dissipating phlegm and resolving masses. It has been reported that the chemical constituents of T. hemsleyanum are mainly flavonoids, polysaccharides, phenolic acids, terpenoids, steroids, volatile oils, alkaloids and so on. It can exert a broad spectrum of anti-tumor effects through various ways such as inhibiting proliferation, migration and invasion of tumor cells, inducing apoptosis of tumor cells, inhibiting angiogenesis of tumor cells, reversing multidrug resistance of tumor cells and regulating body autoimmunity. On the basis of reviewing relevant literature at home and abroad, this paper intends to systematically sort out the chemical and anti-tumor research of T. hemsleyanum, and in order to provide a new idea for its synergistic anti-tumor effect of multi-component, multi-pathway and multi-target, and finally provide theoretical basis for the research and development and clinical application of new anti-tumor drugs of T. hemsleyanum.

ABSTRACT

Macrophages play an important role in inflammation, and excessive and chronic activation of macrophages leads to systemic inflammatory diseases, such as atherosclerosis and rheumatoid arthritis. In this paper, we explored the anti-inflammatory effect of broussonin E, a novel phenolic compound isolated from the barks ofBroussonetia kanzinoki, and its underlying molecular mechanisms. We discovered that Broussonin E could suppress the LPS-induced pro-inflammatory production in RAW264.7 cells, involving TNF-α, IL-1β, IL-6, COX-2 and iNOS. And broussonin E enhanced the expressions of anti-inflammatory mediators such as IL-10, CD206 and arginase-1 (Arg-1) in LPS-stimulated RAW264.7 cells. Further, we demonstrated that broussonin E inhibited the LPS-stimulated phosphorylation of ERK and p38 MAPK. Moreover, we found that broussonin E could activate janus kinase (JAK) 2, signal transducer and activator of transcription (STAT) 3. Downregulated pro-inflammatory cytokines and upregulated anti-inflammatory factors by broussonin E were abolished by using the inhibitor of JAK2-STAT3 pathway, WP1066. Taken together, our results showed that broussonin E could suppress inflammation by modulating macrophages activation statevia inhibiting the ERK and p38 MAPK and enhancing JAK2-STAT3 signaling pathway, and can be further developed as a promising drug for the treatment of inflammation-related diseases such as atherosclerosis.

ABSTRACT

As a commonly used traditional Chinese medicine,Stellaria dichotoma var. lanceolata has a medicinal history of several hundred years, and been included in the Pharmacopoeia of the People's Republic of China. With a sweet fact and mild war nature, it enters liver and stomach meridians. With effects in clearing deficient heat and eliminating fever in infantile malnu, it has been used to treat such diseases as Yin deficiency fever, consumptive fever due to Yin deficiency and infantile chancre fever, and taken as the raw materials of Wuji Baifeng Wan and other traditional Chinese medicine preparations. In recent years,there have been increasing demands for S. dichotoma var. lanceolata from people. However, its wild resources have been over-excavated for a long time,leading to a serious shortage of wild strains. Furthermore, the quality of artificial S. dichotoma var. lanceolata medicine is far different from that of wild cultivars. Meanwhile,pesticide residues and excessive metal standards due to the irrational use of pesticides and chemical fertilizers in the production process, as well as the large number of counterfeits in the market seriously impact the quality,efficacy and drug safety of S. dichotoma var. lanceolata medicine. Therefore, the non-polluted production technology of S. dichotoma var. lanceolata is of great significance. The non-polluted production technology would be an effective mode for promoting the sound development of S. dichotoma var. lanceolata industry in the future and the key to solve the issues. This article summarizes the environment of suitable production area,plantation method, comprehensive soil improvement, field management and rational fertilization technology. It also proposes that the prevention and control of pollution-free safflower pests and diseases should follow the principle of giving priority to comprehensive prevention. The pollution-free and technical regulation system of S.dichotoma var. lanceolata cultivars is built to produce excellent,high-quality and non-polluted production with low content of pesticide residues and heavy metals,and promote the healthy and sustainable development of the global S.dichotoma var. lanceolata industry.

ABSTRACT

<p><b>AIM</b>To investigate the intraspecific variation of Carthamus tinctorius L. (safflower) and establish foundation for further breeding of safflower germplasm resource and screening the quality correlation genes.</p><p><b>METHODS</b>Amplified fragment length polymorphism (AFLP) was carried out to analyze genetic variation of 28 safflower populations collected in China. Unweighed pair-group method of with arithmetical averages (UPGMA) cluster analysis was used to construct a dendrogram and to estimate the genetic distances among the populations.</p><p><b>RESULTS</b>All populations could be uniquely distinguished using 12 selected primer combinations. Similarity coefficients ranged from 0.48 to 0.96 among the populations. Dendrogram revealed distinct segregation of all the cultivars into three main groups and one midst group.</p><p><b>CONCLUSION</b>Limited genetic diversity exists within the tested 28 collections at intra specific level and AFLP-based phyiogeny was not absolutely consistent with that based on morphological characters may be due to the interaction effect between genotype and environment.</p>
